Для воспроизведения модели атрофии РПЭ использовали 22 кролика породы новозеландский альбинос, которым в один глаз субретинально вводили бевацизумаб или физиологический раствор. Забор СЖ проводили до и через 3 мес после инъекции. Во второй серии эксперимента слезу забирали до и на 3, 14 и 21-е сутки после введения бевацизумаба. В СЖ определяли активность 2-МГ ферментативным методом и концентрацию ЭТ-1 методом иммуноферментного анализа.</p></sec><sec><title>Результаты</title><p>Результаты. В СЖ кроликов активность 2-МГ через 3 мес после введения бевацизумаба оставалась в пределах нормы, а после введения физиологического раствора была повышена в среднем вдвое. Содержание ЭТ-1 в СЖ кроликов на 3-и сутки после введения бевацизумаба достоверно увеличивалось как в оперированном, так и парном глазу более чем в 1,5 раза.</p></sec><sec><title>Заключение</title><p>Заключение. Субретинальное введение бевацизумаба не оказывает длительного повреждающего воздействия на сетчатку в отличие от введения физиологического раствора, которое ведет к повышению активности 2-МГ в слезе. Кратковременное увеличение содержания ЭТ-1 в слезе после введения бевацизумаба может свидетельствовать о повышении тонуса сосудов глаза в этот период. Исследование активности 2-МГ и содержания ЭТ-1 в СЖ может быть использовано для мониторинга локальных метаболических сдвигов при воспроизведении атрофии РПЭ, а также для оценки течения посттрансплантационного процесса и адекватности проводимой терапии.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Purpose</title><p>Purpose. To assess the validity of alpha-2-macroglobulin ( 2-MG) activity and endothelin-1 (ET-1) concentration for the characterization of local metabolic disorders in experimental retinal pigment epithelium atrophy (RPE).</p></sec><sec><title>Material and methods</title><p>Material and methods. To reproduce RPE atrophy, 22 New Zealand Albino rabbits were given a subretinal injection of bevacizumab or saline. Tear fluid was collected before the injection and 3 months after it. In the second series of the experiment, tear fluid was also collected on the 3rd, 7th, 14th, 21st and 28th days after bevacizumab injection. Tear fluid was analyzed for the activity of 2-MG using the fermentation method and for ET-1 concentration by the immunoenzymatic method.</p></sec><sec><title>Results</title><p>Results. 3 months after bevacizumab injection, 2-MG activity in the tear remained normal, while after saline injection it was, on average, twice as high as the initial one. ET-1 concentration showed a significant increase of over 1.5 times on the 3rd day after bevacizumab injection both in the tear of the operated and the contralateral eyes.</p></sec><sec><title>Conclusion</title><p>Conclusion. Subretinal bevacizumab injection had no significant lasting damaging effect on the retina, as opposed to saline injection that led to an increase of 2-MG activity in the tear. A transitory increase of ET-1 concentration in the tears after bevacizumab injection may indicate vascular tone elevation in the eye during this period.
